Summary

The purpose of this project is establishing cell lines permissible to koi herpesvirus (KHV) for developing the isolation method of KHV. We established two primary cell lines from
koi (35-passage koi gill and koi fin); the optimum growth medium was either Leibovitz-15 supplemented with 10% fetal calf serum or a mixture containing 60% Earle’s minimum essential medium, 30% Leibovitz-15, 10% tryptose phosphate and 10% fetal calf serum. The optimum growth temperature was between 20-25oC. Those two cell lines were susceptible to KHV and the cytopathic effect (CPE) appeared in koi gill cell 10-14 days post infection; and 20 days post infection in fin cell. The typical CPE showed cytoplasmic vacuolation and cell lysis. The CPE appeared only at first generation of isolation but it disappeared after the sub-passage of the virus in the same cells.
