Training report on the management and operation of animal bio-safety level 3 laboratory in Japan
Abstract
The team began its itinerary on 2 December, 2007 for acquiring the experiences of management and safe operation of animal bio-safety Level 3 laboratory in Japan, and ended on 9 December, 2007. The team visited the research center for zoonosis control in Hokkaido University and prion disease research center in National Institute of Animal Health. The staffs have not only shared the experiences on HPAI and BSE quarantine inspection, animal health control, management and safe operation of animal bio-safety Level 3 laboratory for each other, but developed the cooperative relationship between Taiwan and Japan on emerging zoonoses issue as well. Their achievements also benefit the amendment of intensification for bio-safety of ABSL-3 laboratory in Animal Health Research Institute.

Non-human primate model used for biomedical and new drug research
Abstract
Since 2003, we have received financial support from National Science Council to establish non-human primate animal model and a bio-safety level 3 (BSL-3) lab for developing new drugs. The BSL-3 lab in our institute has passed the qualification examination by CDC (Taiwan) in 2005. During 2003 to 2007, our researchers with the help from National Taiwan University Hospital have performed monkey quarantine and genetic analysis, indicating that Macaca cyclopis, a native species of Taiwan, is most related to the rhesus monkeys in genetics. Now, we possessed 13 heads of M. facicularis imported from Africa and 14 heads of M. cyclopis. In 2008, these monkeys will offer four studies, including genetic and physiological characters, neuron repair, organ transplantation and bone surgery.

Preparation of Limulus Lysate for Endotoxin Assay
Abstract
LAL (Limulus Amebocyte Lysate) test possesses highly sensitivity and specificity for examining bacterial endotoxin, having been wildly used in the detection of pharmaceutical and medical devices. This project is to use the resource of Tachypleus tridentatus (horseshoe crab) residing in the ocean of Penghu region to develop the LAL test. To perform this plan, we have cooperated with Penghu Marine Biology Research Center (PMBRC), which has done the investigation of the horseshoe crab resource, indicating that there are more than 20,000. This product is the basis of all three LAL test methodologies, including gel-clot, turbidimetric, and chromogenic. We have setup the gel-clot method for LAL test. In 2007, we have finished several goals, including: 1) to establish the bleeding technique that one adult horseshoe crab can produce about 1/10 body weight serum; 2) to establish the procedure of serum components extracted from amebocyte; 3) to establish the standard reaction procedure of endotoxin (LPS) with serum components; 4) to test the sensitivity of LAL about 0.05EU/ml. In the future, we will freeze and dry the product, and test the shelf life.
