Development of bivalent vaccines of iridovirus and nervous necrosis virus in grouper

Abstract

In this project, we selected the field isolates with high virulence and antigenicity of grouper iridovirus (GIV) and nervous necrosis virus (NNV) as the seed virus for vaccination by immersion and injection. Different adjuvant formulas and administration rotes were tested for vaccine efficacy. The safety tests of bivalent vaccines with different formulas of adjuvant demonstrated that the vaccine mixed with PCNE oil adjuvant was better in growth of fish. The vaccine efficacy of vaccine mixed with aluminum adjuvant showed the highest protections against NNV and GIV challenge tests. The relative percent survival (RPS) values of NNV and GIV challenge tests were revealed to 77 and 66.6, respectively. The vaccine efficacy of injection type bivalent vaccine was evaluated by intramuscular injection with 106.2TCID50/mL NNV virus and the result revealed the RPS was 75. In addition, the vaccine efficacy of injection type bivalent vaccine was evaluated by intraperitoneal injection with 107TCID50/mL GIV virus and the result revealed the RPS was 88. These results demonstrated the effective protections of injection and immersion- types bivalent vaccines for vaccination against NNV and GIV in giant grouper (Epinephelus lanceolatus).

Application of rapid fluorescent focus inhibition test (RFFIT) for determining rabies neutralising antibody of wild animals and evaluating rabies vaccine efficacy

Abstract

For understanding efficacy of commercial inactivated rabies vaccines and oral vaccine and the serological surveillance of rabies in wildlife, the rapid fluorescent focus inhibition test (RFFIT) for determining the levels of antibody to rabies virus in serum was introduced. After ferret badgers were vaccinated with each of 4 commercial inactivated rabies vaccines (vaccines A, B, C, and D), prescribed for dog and cat, seroconversion (titer >0.5 IU/mL) in vaccinated ferret badgers were detected at 2 weeks post-vaccination. The seropositive rates of ferret badger groups A, B, C were 83% (5/6) at 6 months post-vaccination in comparison with in the group D, which was decreased over time and all titers were less than 0.5 IU/mL after 4 months post-vaccination. In the efficacy test of oral vaccine, seroconversion rate was 26.7% (4/15) at 2 weeks post-vaccination and 40% (6/15) at 1 month post-vaccination. In gem-faced civet, the neutralizing antibody (titer > 0.5 IU/mL) can be elicited after 2 weeks post-vaccination with no observed adverse effect. Forty-six sera from ferret badger, crab-eating mongoose, gem-faced civet and Formosan macaque were tested by the RFFIT. Two sera of ferret badgers which showed nervous clinical signs and illness were tested positive. One of the ferret badgers died and tested positive by fluorescent antibody test.

Analysis cases of avian influenza virus between January to July, 2015

Abstract

Since 2014, H5 HPAIV belonging to clade 2.3.4.4 has spread out over Eurasia and North America, and multi subtypes of H5N2, H5N3 and H5N8 caused outbreaks in Taiwan in January 2015. Up to July 30, a total of 1030 poultry farms had been submitted and 970 cases was confirmed as H5 AIV, which the number of confirmed goose, duck and chicken farms were 692, 58 and 220, respectively. By subtype of virus, 532, 27 and 235 farms were recognized as H5N2, H5N3 and H5N8, respectively. In addition, 68 farms were co-infected with H5N2 and H5N8, and 75 North American related H5N2 viruses were detected in this outbreak. Until July, there are still novel H5N2 (12) and H5N8 (1) distributing in Yunlin County (7), Changhua County (2), Pingtung County (2), Nantou County (1) and Kaohsiung County (1) including chicken, turkey, duck and goose. Both researches about duck intranasal inoculation of H5N8 in Japan and Korea showed insignificant death but shedding viruses. It should pay more attention to surveillance among domestic ducks.

Report on Attendance of the 115th General Meeting of American Society for Microbiology

Abstract

In 2015, the 115th General Meeting of the American Society for Microbiology held at the New Orleans Ernest N. Morial Convention Center, during May 30th to June 2nd. The four-day meeting showcased the central role of microbes in the biosphere by communicating today's cutting edge science in the diverse areas of microbiology through a wide range of interactive workshops, scientific sessions, poster sessions, and networking events. The scientific sessions provided informative and plentiful issues such as, pathogen-host interactions, diagnostic microbiology, epidemiology, and laboratory establishment for microbial research. It was a pleasure as an attendee of this grand occasion, to present our study of bacterial pathogenicity by poster: The arginine at residue 7 of FimY is required for type 1 fimbrial expression in Salmonella enterica serovar Typhimurium. Learning by connecting and discussing with researchers all around the world was the great inspirational experience in this meeting.
