Studies on Rose-Roepke's Acidified Plate-Antigen Test for Bovine Brucellosis

Rose and Rcepk&s Acidified Plate-Antigen Test (APA) was investigated and compared with sndaxd plate-antigen test (SPA) and complement fixation test on 231 bovine sea, The results sen to 1± very gomising and they axe summariJ as follows:

1 .Prexation of Addifierl Plate Antigen: The antigen used was the Bruceila abortus antigen (Plate methce) prcduced by the Institute for the animal &ucellcsis contol pogran in Taiwan, pH was ad justed with glacial acetic acid by glass ektcde pH meter croa Denpa Co.) It was found that when 0(6 to 0.07 ml of glacial acetic acid was adderito 1m1ofsndaxdBrixei]aplateantigen, thepH ofabout4.0.

2.Of the 231 Sea with SPA titers higher than 1:25 225 (9740%), had SPA titers Summary higher than 1:1(X), Whereas 210 (X9%) had APA titers higher than 1:25.

3.ln comparing the APA and SPA test with the compliment fixation test closer agrement to CF test was sen in agglutination test by the APA method, i,e., 1(6% closer agxement was sen in APA t than in SPA t when aggluatintion titers of higher than 1:1(i) were considered to 1± significant.; 27.73% closer agxement was sen in APA t than in SPA t when agglutination titers of higher than 1 :20Dwere considerd to 1e significant and 54.71% closer agrement was sen in APA test than in SPA test when agglutination titers of higher than 1:4(0 were considered to ke CF psitive in the exreriment).

4.It was found that the agglutination titers of APA test were the same whether the sea were inactivael (56t. 30 mm.) or not.
