Summary

Moncclonal antibxlies(Mabi). have bren widely used in biological researches, medical diagnoses and therapies. The prcxlrction of Mab from ascits by mouse can make a high yield; however, the use of animals for large-scale porluction of Mab is restricel for the animal welfare concerns lii the current trend, many in vitro alenatives have been developed for Mab prcduction. in this study the hybridoma cell culture was performed in a 5(Y) niL spinner basket containing 6 g non-woven pcJyestr fabric (NWPF) micrcxarrier. Cells for incculum were prepared at density lx 105
Summary cell.mL-1 of 430 mL c-trite medium. Cells were growing in th culture until day 9 when the perfusion process was initia± Afker S more days, culture was discontinued, During the 14 days culture rericd, four lies surernatant was harvestul in total. ELLSA was applied to evaluat the activity of secreting moncdonal anti&dy (Mab) from culture . According to the result, the Mab activity from spinner basket culture was 2-4 times higher than T-flask culture in control, and the Mab yield of operation was equivalent to 161) Ti 75 flasks. Application of NWPF maturial could lea promising alenative to Mab prcduction while optimal culture condition was established.
