Summary

Protein A-Colloidal gold immunoelectron microscopy (PAGIEM) has been employed to specifically detect Hog cholera virus with swine anti-A?6 serum. This rapid and sensitive immunoassay was found to be applicable to partially purified virus preparation.

Hog cholera virus antigen was labeled in Lit. White embedded ultrathin of Summary CPK cell, using protein A-colloidal gold. Antigens in the rough endoplasmic reticulum. lysosomelike particles, or cvloplasmic vacuole were tagged with 15 nm gold particles.

Working with different viruses and antisera, protein A-Colloidal gold, can be used as an universal gold stain instead of primary or secondary antibodies adsorbed to gold, without losing sensitivity or increasing background.
